Willows and poplars, members of the Salicaceae family, typically accumulate large amounts of phenolic compounds, including benzenoids and salicinoids. P. dividiana is called the Korean aspen and grows in Northeast Asia. No transcriptomic analyses of P. dividiana have been performed.
Result:
In this study, a full-plate 454 pyrosequencing run produced 1,545,322 high-quality reads with an average sequence length of 540 bases. De novo assembly generated 57,322 unique sequences, including 15,217 contigs and 42,105 singletons. Approximately 10,283 (17.9%) unique sequences were annotated using BLAST similarity searches. Six putative genes involved in benzenoid biosynthesis, which starts with phenylalanine, were isolated. Real-time PCR (qPCR) analysis revealed that the mRNA accumulation of the selected 6 genes was clearly enhanced by elicitor (methyl jasmonate) and dehydration (polyethylene glycol) treatments. Salicylic acid, benzoic acid, and salicin were quantified by GC/MS analysis of extracts of leaves and plants of P. dividiana. The salicylic acid content was enhanced by elicitor and dehydration treatment, but the salicin content was reduced.
Conclusions:
The benzenoid biosynthesis pathway genes identified by transcriptomic approaches will help further research studying the functional genomics of P. dividiana for salicin and salicylic acid biosynthesis.
